Granisetron is an antiemetic agent to prevent the nausea and vomiting in conjunction with cancer chemotherapy or with radiation therapy, by blocking 5-HT3 receptors without effect on other receptors such as dopamine D2 receptor and 5-HT4 receptor. The hydrochloride salt of granisetron is a white to off-white crystalline powder; soluble in water and saline; administered intravenously. Chemical designation is endo-N-(9-methyl-9- azabicyclo [3.3.1] non-3-yl)-1-methyl-1H- indazole-3-carboxamide hydrochloride. Ondansetron is another 5-HT3 receptors antagonist used as an antiemetic or antinauseant. The chemical designation is 2,3-dihydro (or 1,2,3,9-tetrahydro)-9-methyl-3-[(2-methyl-1H-imidazol-1-yl) methyl]- 4H-carbazol-4-one. The medicinal actions of granisetron and ondansetron are almost same. Alosetron, 2,3,4,5-tetrahydro-5- methyl- 2-[ (5-methyl-1H- imidazol-4-yl) methyl]-1H-pyrido[4,3-b] indol-1-one, is an 5-HT3 receptor also, but it is used in the treatment of diarrhea-predominant (IBS-D). | ||
